He's charged with sex trafficking, racketeering conspiracy and transportation to engage in prostitution.
Combs denies the charges, but if found guilty, faces life in prison.
CNN's Laura Coates says in opening statements, prosecutors claimed the musician 'ran a criminal enterprise'.
She says opening statements are the moment the prosecution will lay out their case - what they will allege and hope the jurors will hold onto in the eight to 10 week trial.
Court's adjourned for the day after hearing from the first two witnesses.
